Yeast cold-shock microtubule depolymerization
This application note shows how to depolymerize S. pombe microtubules thanks to CherryTemp precise temperature shifts .
S. pombe microtubules were depolymerized by cold shock by shifting cells to 5°C. Fission yeast cells were grown at 25°C and placed on a CherryTemp mounting system for live imaging. Cells were shifted from 25°C to 5°C in less than 10 sec, inducing microtubule depolymerization. Microtubulerepolymerization occurred upon shifting back the temperature to 25°C.
Cells were imaged on a Nikon Ti Eclipse spinning disk. Images were acquired every 20 seconds. Microtubules were visualized withgamma-tubulin GFP
