Agar pad thermalization using CherryTemp
An agar pad (1.5%, thickness 1mm, standard preparation) was mounted in a sandwich between the CherryTemp chip and a glass coverslip. Temperature shifts between 20°C and 37°C were applied. A thermochromic paper was placed above and under the agar pad to measure temperature shifts. The thermochromic paper changes to green when reaching 37°C. Time of shifts is reduced to less than 8 seconds with agar pads of less than 500µM thickness. This mounting method would allow to heat/cool bacteria, plant and yeast during live cell imaging. Agarose pads are a standard method for live cell imaging allowing cell immobilization.
